Optimal nutrient absorption is not only crucial for the \u2018classic\u2019 nutrients such as energy or proteins but it is also important for maximal efficiency of mineral absorption. Given adequate dietary mineral content, this prevents excess (= unabsorbed) calcium and phosphorus from binding with other nutrients in the intestinal lumen and thus rendering them indigestible.<\/p>\n<p>Calcium and phosphorus are among the least efficiently absorbed nutrients in poultry.<\/p>\n<p>While Ca and P are the most directly regulated by 1,25(OH)\u2082D\u2083, the following minerals can also be impacted:<\/p>\n<p>&nbsp;<\/p>\n<p><img decoding=\"async\" class=\"size-full wp-image-1942 aligncenter\" src=\"https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/Table1.jpg\" alt=\"\" width=\"1431\" height=\"635\" srcset=\"https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/Table1.jpg 1431w, https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/Table1-300x133.jpg 300w, https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/Table1-1024x454.jpg 1024w, https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/Table1-768x341.jpg 768w\" sizes=\"(max-width: 1431px) 100vw, 1431px\" \/><\/p>\n<p><strong>\u00a0<\/strong><strong>\u00a0<\/strong><\/p>\n<p><strong>Immunity<\/strong><\/p>\n<p>Nutrient and mineral absorption is also impaired by oxidative stress and inflammation. Dietary supplementation of 1,25(OH)\u2082D\u2083-glycosides supports gut health by enhancing\u00a0tight junction integrity, reducing gut\/tissue permeability, modulating the microbiota and preventing leaky gut.<\/p>\n<p>The interaction between\u00a01,25(OH)\u2082D\u2083 and\u00a0gut health is extremely important and interlinked with innate and adaptive immunity. 1,25(OH)\u2082D\u2083 binds to the\u00a0vitamin D receptor (VDR), which is expressed in the intestinal epithelial cells, and various immune cells in the gut (e.g. intraepithelial lymphocytes, T- and B-cells present in the lamina propria, dendritic cells and macrophages). This interaction modulates gene transcription, influencing both\u00a0immune tolerance and barrier function by promoting among others the expression of antimicrobial peptides such as cathelicidins and \u03b2-defensins. These substances help control pathogens present\/invading in the gut and bloodstream. Furthermore, 1,25(OH)\u2082D\u2083 upregulates tight junction proteins (e.g. occludin, claudins, ZO-1), thus reducing epithelial permeability. This prevents bacterial translocation and endotoxin leakage from the gut into the lumen or even the circulation. In this way, it promotes mucosal homeostasis and reduces the risk of chronic inflammation, one of the most frequent problems in modern poultry production.<\/p>\n<p>Interaction of 1,25(OH)<sub>2<\/sub>D<sub>3<\/sub> in different immune cells<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-1943\" src=\"https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/table2.jpg\" alt=\"\" width=\"711\" height=\"632\" srcset=\"https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/table2.jpg 711w, https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/table2-300x267.jpg 300w\" sizes=\"(max-width: 711px) 100vw, 711px\" \/><\/p>\n<p>&nbsp;<\/p>\n<p><strong>What is the mechanism?<\/strong><\/p>\n<p><strong>Modulation of Adaptive Immunity<\/strong><\/p>\n<p>In the gut, 1,25(OH)\u2082D\u2083 promotes\u00a0regulatory T cell (Treg)\u00a0differentiation. This differentiation is important for the body to recognise its own cells and non-dangerous substances and thus to avoid to start an immune reaction where it is not necessary. Misregulation of these Treg-cells is for example part of allergies or autoimmune disease. Another action is the suppression of the so called Th1 and Th17 response. These cells are also called T-helper cells and are involved in cell-based immunity and inflammation. By modulation TH1 and Th17 response, secretion of the pro-inflammatory cytokines IL-2, IFN-\u03b3, and IL-17 is reduced. On the other side, it modulates Th2 cells (another group of T-helper cells) to a more tolerant response and enhances expression of IL-10, an anti-inflammatory cytokine.<\/p>\n<p>This immunological shift leads to\u00a0an overall immune modulation by a more targeted inflammatory reaction and increased immune tolerance\u2014a state where the immune system recognizes feed antigens as harmless and avoids unnecessary inflammation.<\/p>\n<p><strong>This balance is crucial for preventing immune overactivation in the gut mucosa, dysbiosis and consequently uncontrolled, chronic inflammation!<\/strong><\/p>\n<p><strong>\u00a0<\/strong><\/p>\n<p><strong>Bacterial Chondronecrosis with Osteomyelitis (BCO)<\/strong><\/p>\n<p>BCO is a common leg disorder in fast-growing broilers, characterized by bacterial infection in the growth plate cartilage, leading to necrosis and osteomyelitis. <em>Staphylococcus aureus<\/em> and <em>Enterococcus<\/em> are the most frequent causative agent. Poor gut integrity and systemic inflammation increase the risk of bacterial translocation and subsequent infection in bones.<\/p>\n<p>By improving gut barrier function and reducing systemic inflammation, 1,25(OH)\u2082D\u2083 lowers the likelihood of bacterial \u00a0passage into the bloodstream. Additionally, its role in calcium and phosphorus metabolism enhances bone mineralization, which strengthens skeletal structure and reduces microfractures where bacteria can colonize. Experimental studies done at the University of Arkansas, USA, suggest that dietary supplementation with 1,25(OH)\u2082D\u2083 reduces BCO incidence, improves bone integrity, and supports locomotor health in broilers.<\/p>\n<p>&nbsp;<\/p>\n<p><strong>Panbonis\u00ae,<\/strong> a plant-based feed material which contains glycosylated 1,25-dihydroxycholecalciferol, supports the<img loading=\"lazy\" decoding=\"async\" class=\"wp-image-1945 alignright\" src=\"https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/200825_Herbonis-068-1-scaled.jpg\" alt=\"\" width=\"237\" height=\"356\" srcset=\"https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/200825_Herbonis-068-1-scaled.jpg 1707w, https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/200825_Herbonis-068-1-200x300.jpg 200w, https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/200825_Herbonis-068-1-683x1024.jpg 683w, https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/200825_Herbonis-068-1-768x1152.jpg 768w, https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/200825_Herbonis-068-1-1024x1536.jpg 1024w, https:\/\/plmea.com\/wp-content\/uploads\/2025\/11\/200825_Herbonis-068-1-1365x2048.jpg 1365w\" sizes=\"(max-width: 237px) 100vw, 237px\" \/> vitamin D and calcium and phosphorus homeostasis. It has additionally been shown to increase anti-inflammatory cytokines IL-10 in jejunum and spleen in LPS challenged broilers (Figure 1) and has shown a potent\u00a0epithelium barrier function against the entry of bacteria typical responsible for BCO. Its supplementation can improve gut integrity, modulate immune responses, and significantly reduce the risk of BCO.
